Recent Advances in the Chemistry and Pharmacology of 2-Pyridone Scaffolds

Heterocycles play an important role in bioactive libraries and the pharmaceutical industry. They play an important role in our biological system, so researchers are still designing and discovering novel pharmacologically active molecules that may help to solve recent health issues. Nitrogenheterocycles are a common nucleus in many bioactive natural products and FDA-approved drugs, and they provide an important framework in drug discovery and agrochemical industries. Among all the common Nitrogen-heterocycles, 2-pyridone, a six-membered nitrogen ring, has piqued the interest of researchers due to its potent biological actions such as antibacterial, antitumor, antiinflammatory, antiproliferative, anti-hepatitis B virus, SARS-CoV-2 main protease inhibitor, and analgesic properties. According to a recent review, 2-pyridone structural motifs are important in the synthesis of alkaloids, which are an important class of natural organic compounds.

A Medicinal Chemist′s Guide to Asymmetric Organocatalysis

The majority of drugs work by interacting with chiral counterparts, such as proteins, and we are all too aware of how chirality can have a negative impact on the outcome of a therapeutic regimen. The market for chiral, non-racemic drugs is growing, and it is becoming increasingly important to prepare these compounds in a safe, cost-effective, and environmentally sustainable manner. Asymmetric organocatalysis has a long history, but its renaissance period began only in the first millennium. Since then, this field has advanced to an extraordinary level, as evidenced by the awarding of the 2021 Nobel Prize in Chemistry. We would like to highlight the use of organocatalysis in the synthesis of enantio-enriched molecules that may be of interest to the pharmaceutical industry and the medicinal chemistry community in this review.
